Objectives

  1. Salvia fruticosa Functional Genomics: sequencing of 2,304 ESTs from Salvia fruticosa leaves’ trichomes.

    As an initial step towards understanding the secondary metabolism in Salvia fruticosa trichomes, an EST analysis was performed in a cDNA library constructed from Salvia fruticosa trichomes. A total of 2,304 clones were sequenced, but the exclusion of sequences shorter than 100 bp resulted in 1615 high-quality ESTs with a mean length of 592 bp. Cluster analysis indicated the presence of 197 contigs (908 clones) and 707 singletons, generating a total of 904 unique sequences. Of the 904 unique ESTs, 628 (69.5%) had significant hits in the non-redundant protein database and were annotated.

  2. Cistus creticus Functional Genomics: sequencing of 2,743 ESTs from Cistus creticus subsp. creticus leaves’ trichomes.

    A cDNA library was constructed enriched in transcripts from trichomes isolated from Cistus creticus subsp. creticus young leaves, which are characterized by high levels of labdane-type diterpenes. Initially, 2,743 library clones were randomly selected for single pass sequencing and subsequent vector sequence trimming resulted in 2,022 clones of high-quality sequence. Furthermore, sequences were clustered into 296 contigs and 1,071 singletons. A significant proportion of the ESTs (38%) displayed no significant similarity to any other DNA deposited in databases, indicating a yet unknown function.
